Animals were immunized with a synthetic peptide with the sequence IMVKCWMIDADSRPKF corresponding to peptides 946 to 961 of the HER 1 oncogene (formerly 366 to 381 of the v-erb-B oncogene). In addition, minor activities with proteins of 50-60 kd are detected in these cells as well as a number of additional cell lines and tissues. The IgG1 reactivity of the antibody with the corresponding peptide was confirmed by ELISA. IgG1 was strongly positive while IgG2b was only slightly positive. |

Spleen cells were fused with Sp2/0 myeloma cells. In immunoblot tests, the antibody detects proteins of 170 kd and 150 kd in A431 cells. In addition, minor activities with proteins of 50-60 kd are detected in these cells as well as a number of additional cell lines and tissues. In ELISA tests, the antibody does not detect the homologous region of fes, mos, src, yes, mil, raf, rel, fms, abl, ros, or human insulin receptor. The fgr peptide conjugated to keyhole limpet hemocyanin, however, is recognized by the monoclonal antibody. The IgG1 reactivity of the antibody with the corresponding peptide was confirmed by ELISA. IgG1 was strongly positive while IgG2b was only slightly positive. |

Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density) Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10 exp5 viable cells/ml. Maintain cell density between 1 X 10 exp5 and 1 X 10 exp6 viable cells/ml. |
