Animals were immunized with a synthetic peptide with the sequence LGSGAFGTIYKG(C) corresponding to residues 138-149 of the v-erb B oncogene. Spleen cells were fused with Sp2/0 myeloma cells. In ELISA tests using synthetic peptides representing erb B, fes/fps, fgr, fms, src/yes, mil/raf, abl, mos, CDC28 and the catalytic and regulatory subunit of cyclic AMP dependent protein kinase, the antibody recognizes only the erb B sequence. The 170 kd EGF receptor is weakly detected in immunoblot screening of A431 cells. In addition, a protein of approximately 100 kd is detected in extracts of human brain. The IgG1 and IgG2b reactivities of the antibody with the v-erb B peptide (138 to 149) was confirmed by ELISA. IgG1 was strongly positive while IgG2b was negative. |
