Animals were immunized with liposomes containing high amounts of cholesterol and lipid A as an adjuvant. Spleen cells were fused with P3X63Ag8 myeloma cells. The antibody detects cholesterol in liposomes containing 71% cholesterol but not with liposomes containing 43% cholesterol. It reacts with crystalline cholesterol in a solid-phase enzyme-linked immunosorbent assay. The antibody is capable of detecting cholesterol in situ in the membranes of skeletal muscle sections. A feeder layer of mouse peritoneal macrophages or mouse fibroblasts (see ATCC 56-X, irradiated STO cells) is require to propagate this cell line. |
