Karyotype |
modal chromosome number = 67; range = 59 to 71The stemline modal chromosome number is 67 occurring at 36%, and polyploidy at 0.6%. Fourteen marker chromosomes were common to these cells: 3q+, 11q+, i(13q), t(9q21q), i(17q), i(21q) and 8 others of unknown origin. Additional markers: der(5)t(5;7)(q31;p11) and 1p were found that were specific to a minor subpopulation. Microchromosomes (larger than double minutes) and double minutes (DM) were also found in a small population of cells. Normal N13 was absent; and N21 was found only in the minor subpopulation. One, or occasionally two, Y chromosomes were present in each cell. |
Clinical Data |
68 years adult Caucasian male |
Tumorigenic |
Yes |
Effects |
Yes, within 21 days at 100% frequency (5/5) in nude mice inoculated subcutaneously with 107 cells. |
Complete Growth Medium |
The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.
|
Subculturing |
Remove medium, and rinse with 0.25% trypsin, 0.03% EDTA solution. Remove the solution and add an additional 1 to 2 mL of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37°C) until the cells detach. Add fresh culture medium, aspirate and dispense into new culture flasks.
Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:8 is recommended
Medium Renewal: 2 to 3 times per week |
