Applications |
VacA is a secreted protein that induces vacuolation of epithelial cells. The antibody reacts with purified native s1-m1 VacA but not with denatured VacA. The antibody recognizes a conformational epitope in the carboxy terminal portion of VacA (amino acids 685 to 821). The antibody can be used for ELISA and immunoprecipitation applications. |
Derivation |
Animals were immunized with s1-m1 purified vacuolating cytotoxin (VacA) from Helicobacter pylori strain 60190 (ATCC 49503). VacA is a secreted protein that induces vacuolation of epithelial cells. Spleen cells were fused with Sp2/0-Ag14 cells. The antibody reacts with purified native s1-m1 VacA but not with denatured VacA. The antibody recognizes a conformational epitope in the carboxy terminal portion of VacA (amino acids 685 to 821). It does not recognize type s2-m2 VacA. |

Complete Growth Medium |
The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
|
Subculturing |
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density) Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 10 exp5 viable cells/ml. Maintain cell density between 2 X 10 exp5 and 2 X 10 exp6 viable cells/ml. |
Cryopreservation |
culture medium 95%; DMSO, 5% |
