Insert contains internal PstI (1), TaqI (2), MboII (2), HhaI (1), HinfI (2), HindIII (1), XbaI (1), and HincII (1) sites. Restriction digests of the clone give the following sizes (kb): PstI--4.4, 0.9; EcoRI/NdeI--3.0, 2.3; EcoRI--5.3; HindIII--3.8, 1.5; BamHI--5.3. The insert is from the 3' half of the cDNA. No HaeIII is known in the cDNA of the albumin gene, so the variants must be intronic. In one variant, 2 of 42 or f=0.05 were missing the 2.7 and 1.6 kb bands. In another variant, bands 4, 2.7, 0.7, 0.65, 0.6, and 0.55 were missing (f= 0.21). THe second variant has been validated in family studies. When DNA is digested with HaeIII, the probe detects bands at 4, 3.4, 2.9, 2.7, 1.6, 0.95, 0.75, 0.72, 0.65, and 0.6 kb. |
