Restriction digests of the clone give the following sizes (kb): BamHI--4.5, 2.8; BstEII--7.4; EcoRI--4.9, 2.4; NotI--7.4; SacI--7.0, 0.44. Digestion with EcoRI produces fragments of 5.0 and 2.4 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes a chimera in which aa 1-48 of processed human factor VII (Gla domain) replace the corresponding region of factor IX. |
